What was filed
Tanabe Pharma filed loncastuximab tesirine (MT-2111), an anti-CD19 antibody-drug conjugate, for relapsed or refractory diffuse large B-cell lymphoma. The filing uses a Japanese Phase I/II study and overseas data.
Read the denominator
The Japanese Phase II portion was an open-label, single-arm study in 43 patients. Independent central review found an objective response rate of 58.1% (25/43) and complete response rate of 41.9% (18/43). Median duration of response was not reached at 15.21 months of median follow-up. The small, uncontrolled, company-reported dataset does not establish superiority.
Overseas approval is not Japanese approval
In the United States the drug has accelerated approval for adults with relapsed or refractory large B-cell lymphoma after at least two systemic treatment lines. Continued approval may depend on confirmatory benefit. This does not establish Japanese access, price or prescribing.
Important risks
The US label warns about effusion and edema, myelosuppression, infection, hepatotoxicity, cutaneous reactions and photosensitivity, and embryo-fetal toxicity. A future Japanese label, if approved, may differ.
International-patient relevance
Useful records include pathology, CD19 status, prior treatment lines, transplant or CAR-T history, imaging, organ function and serious adverse events. The current filing does not support promises of access in Japan.
