Large-scale centromere analysis with existing WGS data: a research method, not a clinical test

The study analyzed 3,332 genomes and 142 oligodendroglioma cases to explore centromere diversity and formation of the 1p/19q co-deletion.

What changed

The team developed ascairn, which uses 27-base rare k-mers to infer centromere types from existing short-read whole-genome sequencing data. Median inference accuracy in the performance assessment was 92.7%.

Cancer relevance

In 142 oligodendroglioma cases, breakpoints involving chromosomes 1 and 19 were mapped within centromeres. The analysis suggests that structural variation between individuals may be related to formation of the characteristic 1p/19q co-deletion.

Evidence boundary

Status: basic research / genomic method. Ascairn is not an approved clinical test or treatment, and the publication does not establish immediate utility for individual treatment decisions.

Long-term value

The method may enable researchers to revisit large short-read WGS collections and study repetitive genomic regions that were previously difficult to analyze. Clinical validation remains future work.
