How it works
An L-DOPA-bearing, photoresponsive polymer preferentially entered cancer cells. In acidic endosomes it accumulated near the membrane; illumination then disrupted the membrane and released the co-delivered mRNA carrier into the cytoplasm.
Results
Protein production increased more than 80-fold after illumination in mouse colon-cancer and human cervical-cancer cells. No production was observed in non-cancer mouse fibroblasts under the same conditions. Expression was also localized to illuminated sites in a mouse tumor model.
What remains unknown
This is a delivery-control proof of concept, not evidence that human cancer has been treated. Therapeutic benefit, normal-tissue effects, immune responses, repeat dosing and deep-tissue illumination all require further study.
