CheckMate 648: five-year follow-up of nivolumab combinations in esophageal cancer

The five-year analysis supports durability of established standard regimens; it is not a new approval.

Overall-survival benefit persisted at five years

Among 970 untreated patients with unresectable advanced, recurrent or metastatic ESCC, both nivolumab-containing regimens maintained an overall-survival advantage versus chemotherapy alone.

Five-year survival in tumor-cell PD-L1 ≥1%

Nivolumab + chemotherapy 12%; nivolumab + ipilimumab 18%; chemotherapy 7%.

PFS differed by regimen

Nivolumab + chemotherapy improved progression-free survival, while nivolumab + ipilimumab did not show a PFS improvement versus chemotherapy.
